Would the format of the drive matter?

Yup, it should be FAT32, but you can get NTFS to work.

Use a branded pendrive and one which eats less power and mostly it has to be formatted to FAT or FAT32 not sure about filesystem :-/

I bought a 64Gb Sandisk from Amazon (UK) (make sure to buy direct from Amazon, not another seller!)
Default comes formatted as exFAT, so PC didn't recognise until formatted with FAT32.
You can either format thru your phone or on PC as I recall.
Depending on your ROM, I would suggest you will be better off with FAT32. (Windows doesn't recognise exFAT anyway).

USB extension for?...
Android itself supports USB hardware (such as mice and keyboard if you're referring to that.) But stock ROM and other custom ROMs for the Nook Tablet do not support it. In other words, no.

OK. Well the kernel for android supports it. But I know the stock ROM doesn't support it. I'm not sure if there are any custom roms out there who's kernel supports USB keyboards.
